Using a Flatbed Scanner for Three Dimensional Reconstruction

The chicken was then frozen and sliced up to 3-mm sections with a commercial meat slicer. The slices were then put on letter-size overhead transparency plastic sheets. These were then placed on a flatbed scanner and, in order of the sections, scanned. The scans were saved as bitmap files and adjusted for brightness and contrast in Adobe Photoshop . The scans were then imported into Corel Draw , and the blanched, ablated area of the chicken was traced in the draw program. These outlines were exported as .DXF outlines and saved. These outlines were then imported into a CAD program (in this case Autocad ). The outlines were extruded into solid objects in the CAD program to a depth of 3 mm. Once the objects were generated in the CAD program, using the program's tools to calculate the volume of the ablation became a simple matter. CAD models, both detailed and rough representative models, can be rendered and then easily trimmed and made into a separate object. Another way to quickly grab an image is to use the key (on a PC). This saves an image on the screen to memory. This can then be pasted in to a bitmap image editing program like Photoshop by using the New file command, then the Paste command. Recording Results

Labeled embryos are mounted in 100 glycerol. Because of the photosensitivity of the yolk cell, embryos at early developmental stages are first treated for 5 min in an acidic buffer (either PBS, pH 3.5, or glycine buffer, pH 2.2). This treatment prevents photoreactivity of the yolk proteins, and even under intense light the yolk cell remains unstained. However, this acidic treatment affects embryo morphology. Therefore, although very convenient at early developmental stages (when only a few structures are formed and when the photolabeling of the yolk is a limiting factor for the observation), this acidic treatment should not be used for embryos older than the 15-somite stage. Embryos are observed in glycerol between the slide and coverslip (using bridges made of four coverslips of thickness 1.5 mm).
